What is Hexarelin?

Hexarelin is a synthetic hexapeptide growth-hormone-releasing peptide (GHRP) studied as an agonist of the growth-hormone-secretagogue receptor 1a (GHS-R1a). Activation couples the receptor through Gq/11 to phospholipase C, generating inositol-1,4,5-trisphosphate and diacylglycerol, mobilizing intracellular calcium and engaging protein kinase C in pituitary somatotroph models - a pathway mechanistically distinct from GHRH-receptor cAMP signaling. Two structural modifications, the D-2-methyl-tryptophan at position 2 and the D-phenylalanine at position 5, confer resistance to enzymatic degradation and shape receptor affinity. Beyond the pituitary, hexarelin is also studied for binding to the scavenger receptor CD36 in cardiovascular-tissue models. All description is in-vitro and ex-vivo receptor pharmacology; no therapeutic or human-use claim is made.
