Reconstitution principle and concentration math

The resulting stock concentration equals peptide mass divided by solvent volume. For example, adding 2 mL of bacteriostatic water to a vial containing 5 mg of AOD-9604 yields a stock of 2.5 mg/mL; adding 1 mL to the same 5 mg vial yields 5 mg/mL. Because AOD-9604 has a defined molecular weight of 1815.06 g/mol, a mass concentration can also be expressed in molar terms for assay work using moles = mass / molecular weight. Choosing the solvent volume is purely a function of the working concentration the assay requires.

Bring the lyophilized AOD-9604 vial and the solvent to room temperature before opening to limit condensation on the dry peptide.

Calculate the solvent volume needed for the target stock concentration using the known vial mass and the relationship concentration = mass / volume.

Wipe both the peptide vial septum and the bacteriostatic-water vial septum with an alcohol wipe.

Draw the calculated volume of bacteriostatic water and add it slowly down the inner wall of the peptide vial, letting it run onto the side rather than directly onto the lyophilized pellet.

Do not shake. Allow the peptide to dissolve undisturbed, with gentle swirling only, until the solution is clear.

Confirm full dissolution visually; a clear, particle-free solution indicates the reference material is in solution and ready for laboratory aliquoting.

Label the reconstituted vial with peptide name, stock concentration, and reconstitution date for traceability.
